In a biuret test, some protein was mixed with sodium hydroxide solution. Which of the following chemicals should be added to the mixture for a positive result?

A. mercurous nitrate

B. copper sulphate

C. mercuric nitrate

D. sodium carbonate




Biuret test is a set up deployed to test the presence of protein in a food substance.

To conduct this test, biuret reagent made up of a mixture of sodium hydroxide and copper sulphate is mixed with the food substance. If protein is present, violet color will be observed.

  2. B is correct. 1% of copper sulphate mixed with sodium hydroxide makes up biuret reagent.
